چکیده
Dear editor; After receiving the comments from the readers, we checked the results carefully and decided to repeat all the experiments. As the first author has already left the lab, other members are still trying to repeat these experiments. Because it takes a long time to have these experiments redone, we decide to retract our paper entitled “Stable chloroplast transformation of immature scutella and inflorescences in wheat (Triticum aestivum L.)” by Cuiju Cui, Fei Song, Yi Tan, Xuan Zhou, Wen Zhao, Fengyun Ma, Yunyi Liu, Javeed Hussain, Yuesheng Wang, Guangxiao Yang, and Guangyuan He, published in Acta Biochimica et Biophysica Sinica, Volume 43, Number 4, April 18, 2011, pp. 284-291 (DOI: 10.1093/ abbs/gmr008), until all the results are reconfirmed. We would like to thank the readers for their critical comments and we feel sorry to the readers for the inconveniences caused. Acta Biochim Biophys Sin 2012, 44: 373 | a The Author 2012. Stable chloroplast transformation of immature scutella and inflorescences in wheat (Triticum aestivum L.).
Chloroplast transformation in wheat was achieved by bombardment of scutella from immature embryos and immature inflorescences, respectively. A wheat chloroplast site-specific expression vector, pBAGNRK, was constructed by placing an expression cassette containing neomycin phosphotransferase II (nptII) and green fluorescent protein (gfp) as selection and reporter genes, respectively, in the inte...
متن کاملEnhancement of Agrobacterium-mediated transformation efficiency in immature embryo of Triticum aestivum, cv. Arya
An efficient Agrobacterium-mediate transformation method was developed by employing different duration of sonication, Agrobacterium strains, type of inoculation medium and concentrations of acetosyringone in Arya cultivar of wheat. Immature embryos were used as an explant for inoculation with Agrobacterium tumefaciens harboring the recombinant pBI121 plasmid. Among the durations of sonication t...
متن کاملAgrobacterium-mediated transformation of mature embryos of Triticum aestivum and Triticum durum
Plant regeneration studies in cereals have been undertaken in immature embryos, scutellum and also in immature inflorescence tissue. The wheat mature embryos can also be employed for callusing and regeneration, as they are available throughout the year and have presently been employed for transformation studies. An efficient and reproducible method for Agrobacterium-mediated transformation of m...
متن کاملBiolistic transformation of wheat using the HMW-GS 1Dx5 gene without selectable markers.
Genetic manipulation using linear DNA was applied to the common wheat variety Xindong No. 26 via particle bombardment with the aim to improve bread-making quality of flour. Initially, 2 biolistic parameters, helium pressure and target distance, were optimized using plasmid pAHC25. We transformed wheat immature embryo scutella with the linear 1Dx5 gene without selectable markers. The highest tra...
متن کاملThe Effect of Priming and Salinity on Physiological and Chemical Characteristics of Wheat (Triticum aestivum L.)
In order to study of the effect of priming and salinity on physiological and chemical characteristics of wheat(Triticum aestivum L.), an experiment was carried out at the Experimental Farm of Shiraz University. Results showedthat primed plants significantly reduced its gas exchanges by accelerating senescence under a series of salt stress, which became more serious along with the increasing of s...
